ABSTRACT

The Covid-19 virus was discovered in Wuhan, China in 2019, and spread globally, prompting the WHO to declare the existence of this virus as a pandemic outbreak. Indonesia became one of the countries affected by the entry of this virus in March 2020, and an economic sector including tourism experienced paralysis. There was a decrease in the number of tourists which resulted in a low percentage of hotel occupancy. This study aims to determine the effectiveness of the Eastparc Hotel strategy in responding to the staycation trend towards occupancy rates during the Covid-19 pandemic. The method used in this study is a qualitative description method with data acquisition in the form of primary data which is collected after in-depth interviews with the HR Director of Eastparc Hotel and secondary data derived from literature studies through articles, theories, and dictates. The results showed that the Eastparc Hotel was greatly affected by the pandemic. There was a drastic decline in hotel occupancy and even a temporary suspension of operations took effect. However, Eastparc Hotel is being adaptive by bringing staycation innovation into the concept of facility development. The hotel is also always consistent with the principles of healthy and cleanliness in serving and providing facilities. Both of these things are effective in increasing hotel occupancy, in the sense that Eastparc Hotel has implemented an effective strategy to survive a pandemic. Furthermore, development activities are continued to be carried out of the flow of digitalization to improve the sustainability of the Eastparc Hotel.

ABSTRACT

Background: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been associated with immune hyperactivation and high levels of proinflammatory cytokines. Extensive lung infiltration by CD169+ inflammatory monocytes and presence of activated CD169+ alveolar macrophages suggest monocyte/macrophages are key drivers of severe morbidity and mortality. In this study, we determined whether CD169 mediated ACE2-independent SARS-CoV-2 entry and restricted viral genome replication in macrophages triggers pro-inflammatory cytokine expression. Methods: Monocyte-derived macrophages (MDMs) and PMA-differentiated THP-1 macrophages engineered to constitutively express CD169, ACE2, or CD169 and ACE2 were infected with USA-WA1/2020/SARS-CoV-2 isolate with or without Remdesivir pre-treatment. To identify mechanism of innate immune activation, nucleic acid sensing pathways were selectively depleted in CD169+ macrophages. Extent of viral genomic (gRNA) and sub-genomic (sgRNA) expression and induction of pro-inflammatory cytokines was determined by qRT-PCR and single molecule RNA FISH analysis. Viral protein expression and infectious virus particle production was determined by immunofluorescence analysis and TCID50. Results: While productive virus infection (viral protein expression and infectious virus particle release) was only observed in ACE2+ macrophages, SARS-CoV-2 N or S expression and infectious virus production was not observed in CD169+ macrophages. Co-expression of ACE2 and CD169 significantly enhanced infectious virus production and spread. Interestingly, smFISH and RT-qPCR analysis revealed CD169+ cells express cytosolic negative-strand gRNA and positive strand sgRNA. Importantly, CD169-mediated SARS-CoV-2 infection of macrophages and expression of viral mRNAs led to induction of pro-inflammatory cytokines, IL-6, TNFα, and IL-1β, despite lack of viral protein expression in CD169+ macrophages. Pre-treatment with Remdesivir blocked de novo expression of viral mRNAs and induction of inflammatory cytokines in CD169-dependent infection of macrophages. Furthermore, knockdown of cytosolic RLRs (RIG-I and MDA-5) or MAVS significantly attenuated inflammatory cytokine expression in CD169+ macrophages, confirming that nucleic acid sensing of restricted cytosolic viral mRNA expression in macrophages triggers innate immune activation. Conclusion: These results suggest that restricted SARS-CoV-2 infection of CD169+ macrophages contributes to COVID-19-associated hyperinflammatory cytokine response.

ABSTRACT

One of the basic problems experienced by teachers, parents, and students due to the impact of COVID-19 is the adaptation of learning methods for students which requires the use of technology as a learning medium. This study aims to describe the use of Quipper School in learning children's literature in the era of the COVID-19 pandemic to increase the reading interest of elementary school students. The method used in this study is a qualitative method, this method examines a group of people, a set of conditions or a class of events which is then analyzed to interpret the data obtained by solving problems and a relevant final phenomenon. This research will provide an overview of the problems or phenomena that occur in the Laboratory Elementary School, State University of Malang. The informants in this study were 47 students. The data collection techniques were carried out by means of observation, interviews and documentation. The findings obtained in this study are empirical data regarding the use of Quipper School in learning children's literature in the era of the COVID-19 pandemic to increase interest in reading. The use of Quipper School has a positive impact on increasing students' interest in reading. The increase in students' reading interest is shown by understanding the content of the text in each material presented, better independent reading activities, critical thinking skills in responding to the content of the text. © 2022 IEEE.

ABSTRACT

BACKGROUND: Coronavirus disease-19 (COVID-19) has been declared a world pandemic by World Health Organization (WHO). The ability of COVID-19 to be transmitted quickly causes fear and excessive worry in certain groups of people, such as the elderly and those with chronic diseases, including oncology and gynecology patients and the caregivers around them (family members). AIM: This study investigates knowledge, attitude, and negative emotions among gynecologic oncology patients and their families during the COVID-19 pandemic at Hasan Sadikin Public Hospital. METHODS: In this descriptive study, 100 patients with gynecologic malignancies at Hasan Sadikin Hospital, Bandung, Indonesia, were assessed in May 2020–April 2020, who asked for their current knowledge, attitude, as well as negative emotions through Depression Anxiety Stress Scale (DASS-42). RESULTS: The average age of the respondents in this study was 45.50. There were 31 respondents with low-level knowledge, 36 with moderate-level knowledge, and 33 with a high level of knowledge about COVID-19. Based on the attitude assessment, only 44 respondents (44%) always wore masks, 54 (54%) kept a distance of 1 meter, and 42 (42%) kept away from the crowd. An evaluation using the DASS-42 showed that 3, 4, and 0 subjects experienced severe depression, severe anxiety, and severe stress, respectively. CONCLUSIONS: All Indonesian citizens must take necessary measures to prevent COVID-19, including wearing masks, maintaining a minimum distance of 1 m, and washing hands. This study found that not all respondents could comply with the health protocols that were carried out to prevent the transmission of COVID-19.
